How I did it: I finally got a contact at the ASPCA, submitted my resume and got an interview!  I was actually not selected for the job and was absolutely devastated.  After 2 years of trying to get an interview, when I finally got one and was told DURING the interview that I was not right for the position, I was so dejected.  But a second job was up on their website a few months later, and I used the same contact to apply for that job.  I got the interview, and about an hour after I interviewed, I was called and offered the job! I was so happy and still am.

Lessons & tips: Use whatever networking contacts you can, and don't give up hope even when you think it's lost.
