It took 324 licks at one spot to get to the Tootsie Roll Center. This means that as soon as a breakthrough to the center was made, it was over.
Now, there are other ways to calculate it:
1) All of the sucker material is gone.
2) Lick until the Tootsie Roll stuff is also gone.
