Not so sure about an entire week; it just depends on the destination. Andrea and I went to New Orleans, and had seen all we wanted to see in 2.5 days. Although over the course of the year, this might be feasible, maybe just not an entire week in one city.
How to spend 1 week+ every year somewhere I've never been
How I did it: Each year we budget, save and plan a trip to somewhere new and wonderful. We've done this each year since 2004, and don't intent to stop. Since we don't just sign up for tours, our international trips might be a little time consuming to plan, but so incredibly worth it. In previous years we've visited
04: New Mexico/Colorado + PA /WV
05: Germany
06: California + Montana/Wyoming
07: Italy
08: New Zealand / Sydney AUS.
This year? Washington/Oregon + Peru. and beyond? Oh, there's a very tanative list. Don't worry
Lessons & tips: Really seeing places takes planning. Sure it can be fun to hop in a car/ on a plane and just go, but if you want to most bang for hour buck as well as time, planning is essential.
Resources: Maps and directions are always important. Between Michelin, Google, and Mapquest, you can ususally get custom directions to whereever you need to go.
DK's eyewitness guide books are great for planning, and extra details can be found through frommer's websites. also, staying in hostels (hostelworld.com) is by far the cheapest and easiest way to go outside the US.

I do it every year-it’s great. Sometimes I like it, sometimes I don’t.
I have 3 days, spent in New York.
Haven’t decided where I’m going this summer, but that will be over a week.
